What is Beta-Alanine, and How Does it Work in the Body?

Beta-alanine is a non-essential, natural amino acid. In the muscle, beta-alanine is the rate-limiting precursor to carnosine. Carnosine acts as an acid buffer within the muscles during training. (1) 

During sustained or repeated high-intensity exercise, anaerobic glycolysis contributes substantially to ATP production. This is associated with an accumulation of hydrogen ions (H+), which lowers muscle pH and can contribute to fatigue and reduced muscle force. Carnosine helps buffer these changes in muscle acidity, which is one reason beta-alanine may benefit high-intensity exercise performance. (2) 

Carnosine is an important buffering agent, the first line of defense against the muscles becoming too acidic. You can’t make carnosine without beta-alanine, and that’s where supplementation comes into the picture. By increasing beta-alanine, you increase carnosine, and therefore the buffering capacity of the muscles. (2)

Beta-alanine supplementation is associated with delayed fatigue and improved muscle function during high-intensity training, including weightlifting, where high-power lifts increase muscle acidity. (1)

What Causes the Tingling? The Science of Paresthesia Explained

The pre-workout itch or tingling sensation is one of the unexpected effects of beta-alanine. Clinically, this is called skin paresthesia, an unpleasant tingling or itching sensation caused by activation of sensory neurons in the skin. 

Scientists have discovered that paresthesia from beta-alanine is not a histamine reaction, but the exact mechanisms remain elusive. While the tingling can be uncomfortable, it is harmless. Beta-alanine has a high safety profile at the researched dosages. (3, 4)

One randomized controlled trial showed that participants who received beta-alanine instead of placebo experienced increased perceived pain afterword. Interestingly, they also experienced more “love and care of nature” during outdoor exercise. (5) 

Bottom line: Does beta-alanine make you tingle? Yes, it might, but the side effect is temporary, safe, and worth the benefits. 

The Clinical Dose for Beta-Alanine: What the Research Actually Recommends 

A 2025 systematic review of 9 studies reports that doses between 4 and 6.4 grams per day offer the most performance benefits. Other research shows benefits at 2- to 6-grams per day. (1, 6)

Further, divided doses seem to offer additional benefits. When the daily dose is divided into smaller doses and taken several times per day, they “were more likely to enhance maximal strength and power-related outcomes.” One evidence-based strategy is to take two to four 1.6-gram doses. (1, 2) 

On the other hand, studies using higher doses or sustained-release formulas didn’t show the same benefits. The cumulative daily dose is the most important factor for improving strength and power adaptations. (1)

Beta-Alanine and Endurance: What the Evidence Says About Performance Benefits 

As a natural pre-workout, beta-alanine is an ergogenic aid. In other words, it helps to enhance physical performance. It’s been studied in dozens of randomized controlled trials. 

In a 2012 meta-analysis of 15 studies, the pooled analysis explored 57 measures of 23 exercises, 18 supplement protocols, and 360 participants. The results suggest that beta-alanine improved exercise performance better than placebo. The calculated performance improvement was 2.85%, which is significant. (2)

In a 2017 systematic review and meta-analysis of 40 double-blind placebo-controlled trials, researchers looked at 65 different exercise protocols, 70 exercise measures, and 1461 participants. They discovered a more nuanced performance benefit that depended on the type of exercise and its duration. Overall, however, there was a significant benefit of beta-alanine on exercise capacity and performance. (7)

A more recent 2023 systematic review of clinical trials on combat sports found similar results. Beta-alanine supplementation improved strength, power, and exercise capacity parameters. (8)

How to Evaluate Whether Your Pre-Workout has a Proper Beta-Alanine Dose 

According to research, the optimal total daily dosage range for beta-alanine is between 2 and 6.4 grams per day. This is the range to look for on the “Supplement Facts” panel of your pre-workout or non-stim pre-workout supplement.
